dc.descriptionIkan selar (Selaroides Leptolepis) merupakan salah satu jenis ikan yang tersebar luas di wilayah perairan Indonesia dan berpotensi menjadi sumber kolagen. Penelitian ini dilakukan untuk menentukan karakteristik kolagen larut asam (ASC) dan kolagen larut pepsin (PSC) dari kulit ikan selar (Selaroides leptolepis). Pada penelitian ini, kolagen diekstraksi dengan menggunakan kulit ikan selar dengan menggunakan metode larut asam dan penambahan enzim pepsin. Proses pretreatment dilakukan dengan menggunakan NaOH 0,1 M selama 18 jam. Sampel kulit ikan selar kemudian diekstraksi dengan menggunakan asam asetat (CH3COOH) konsentrasi 0,5 M selama 12 jam dan 24 jam, serta penambahan enzim pepsin dengan konsentrasi 0%, 1% dan 5%. Hasil kolagen yang diperoleh kemudian dianalisis kadar air dan kadar abu, FTIR (Fourier Transform Infrared Spectrofotometry), analisis spektrofotometer uv-vis dan analisis DSC (Differential Scanning Calorimetry). Dari hasil ekstraksi diperoleh kenaikan rendemen kolagen yang berbanding lurus dengan konsentrasi enzim dan waktu ekstraksi. Kondisi optimum dicapai pada sampel dengan konsentrasi enzim 5% dengan waktu ekstraksi 24 jam dengan rendemen sebesar 18,64 %. Hasil uji air dan uji abu kolagen masing masing diperoleh 2,37% dan 15,87%. Dari hasil FTIR untuk ke 6 variasi sampel menunjukan bahwa proses ekstraksi dengan asam dan pepsin dapat mempertahankan struktur triple helix pada kolagen. Berdasarkan hasil Analisis DSC (Differential Scanning Calorimetry) diperoleh suhu denaturasi kolagen larut pepsin yaitu 64,05°C.en_US
dc.description.abstractSelar fish (Selaroides Leptolepis) is a type of fish that is widely distributed in Indonesian waters and has the potential to be a source of collagen. This study was conducted to determine the characteristics of acid soluble collagen (ASC) and pepsin soluble collagen (PSC) from the skin of selar fish (Selaroides leptolepis). Collagen was extracted by using the skin of selar fish using the acid solution method and the addition of the enzyme pepsin. The pretreatment process was carried out using 0.1 M NaOH for 18 hours. Selar fish skin samples were then extracted using 0.5 M acetic acid (CH3COOH) for 12 hours and 24 hours, as well as the addition of pepsin enzymes with variations in the concentrations of 0%, 1% and 5%. The results of the collagen obtained were then analyzed for moisture content and ash content, FTIR (Fourier Transform Infrared Spectrophotometry), uv-vis spectrophotometer analysis and DSC (Differential Scanning Calorimetry) analysis. From the extraction results, the increase in collagen yield is directly proportional to the enzyme concentration and extraction time. Optimum conditions were achieved in samples with 5% enzyme concentration with extraction time of 24 hours with a value 18.64 % by mass. The results of the water Content and collagen ash Content were 2.37% and 15.87%. From the FTIR results for 6 sample variations, it shows that the extraction process with acid and pepsin can maintain the triple helix structure of collagen. Based on DSC (Differential Scanning Calorimetry) analysis, the denaturation temperature of pepsin-soluble collagen was 64.05°C.en_US
