Collagen Extraction from Yellowscad Fish Skin (Selaroides leptolesis) using Acid Solluble Collagen Extraction Method with the Addition of Papain Enzyme This research is about the utilization of the potential of the Yellowscad fish in Indonesia by extracting collagen from the skin of the Yellowscad fish and adding the papain enzyme to the acid solvent with the aim of increasing the yield of collagen produced when added and not added with papain enzymes and seeing the characteristics of the collagen produced. There are four stages of research, namely sample preparation, pretreatment, extraction process, and salting out. Extraction was carried out with variations in the addition of the enzyme papain 0; 1; and 5% and variations in immersion time of 12 and 24 hours. The results of the study were dry collagen clumps which were then analyzed for ash content, moisture content, and collagen yield and were characterized using FTIR (Fourier Transform Infrared Spectroscopy), UV-Vis (UltraViolet-Visible), and DSC (Differential Scanning Calorimetry) tools. The collagen obtained showed an increase in the yield of collagen along with the addition of papain enzymes. The highest yield was obtained with the addition of 1% papain enzyme with an immersion time of 12 hours with a value of 3.736% mass. The results of the analysis of ash content and water content showed the numbers 35.893% and 3.868%, respectively. The results of FTIR characterization showed 5 complete main groups of collagen from both fractions. The five groups are amide A, amide B, amide I, amide II, and amide III. From the results of the DSC analysis, it was found that the collagen denaturation temperature was 82.29 0Cen_US

Ekstraksi Kolagen dari Kulit Ikan Selar Kuning (Selaroides leptolesis) dengan Metode Ekstraksi Kolagen Larut Asam dengan Penambahan Enzim Papain Penelitian ini tentang pemanfaatan potensi dari ikan selar kuning yang ada di Indonesia dengan mengekstraksi kolagen dari kulit ikan selar kuning serta menambahkan enzim papain pada pelarut asam dengan tujuan untuk meningkatkan rendemen kolagen yang dihasilkan apabila ditambahkan dan tidak ditambahkan dengan enzim papain serta melihat karakteristik kolagen yang dihasilkan. Terdapat empat tahap penelitian, yaitu preparasi sampel, pretreatment, proses ekstraksi, dan salting out. Ekstraksi dilakukan dengan variasi penambahan enzim papain 0; 1; dan 5% serta variasi waktu perendaman 12 dan 24 jam. Hasil penelitian berupa gumpalan kolagen kering yang selanjutnya dianalisis dari kadar abu, kadar air, dan rendemen kolagen serta dilakukan karakterisasi menggunakan alat FTIR (Fourier Transform Infrared Spectroscopy), UV-Vis (UltraViolet-Visible), dan DSC (Differential Scanning Calorimetry). Kolagen yang diperoleh menunjukkan adanya peningkatan rendemen kolagen seiring penambahan enzim papain. Rendemen tertinggi diperoleh pada penambahan enzim papain 1% dengan waktu perendaman 12 jam dengan nilai sebesar 3,736% massa. Hasil analisis kadar abu dan kadar air menunjukkan angka 35,893% dan 3,868%. Hasil karakterisasi FTIR menunjukan 5 gugus utama kolagen yang lengkap dari kedua fraksi. Kelima gugus tersebut yaitu amida A, amida B, amida I, amida II, dan amida III. Dari hasil analisis DSC didapatkan bahwa suhu denaturasi kolagen yaitu 82,29 0C.en_US
